Social media serves as a vast repository of data, offering insights into public perceptions and emotions surrounding significant societal issues. Amid the COVID-19 pandemic, long COVID (formally known as post-COVID-19 condition) has emerged as a chronic health condition, profoundly impacting numerous lives and livelihoods. Given the dynamic nature of long COVID and our evolving understanding of it, effectively capturing people's sentiments and perceptions through social media becomes increasingly crucial. By harnessing the wealth of data available on social platforms, we can better track the evolving narrative surrounding long COVID and the collective efforts to address this pressing issue.

During the 2023-2024 respiratory syncytial virus (RSV) season in the United States, 2 new RSV prevention products were recommended to protect infants in their first RSV season: nirsevimab and Pfizer's maternal RSV vaccine. Postlicensure studies are needed to assess prevention product impact and effectiveness.

Hospitalization is most common for severe pertussis cases and those with serious complications caused by pertussis. In mainland China, the immunization program consists of primary vaccinations at 3, 4, and 5 months, followed by a booster at 18 months with DTaP. It remains uncertain whether the first dose at 3 months, rather than 6 weeks as WHO recommended, may increase disease burden, as delayed immunization may lead to lagged protection. This study aimed to determine pertussis hospitalization rates by active surveillance in China, shedding light on immunization strategies for better disease control.

Bacterial cell factories have been successfully engineered to efficiently assemble spherical polyhydroxybutyrate inclusions coated with functional proteins of interest. In these submicrometer-sized core-shell assemblies, proteins are bioconjugated to the polymer core, enabling bioengineering for uses as bioseparation resins, enzyme carriers, diagnostic reagents, and particulate vaccines. Here, we explore whether these functional protein-polymer assemblies could be restructured via dissolution and subsequent precipitation while retaining the functionality of the conjugated protein. Polymer core-protein shell assemblies were completely dissolved in chloroform. Subsequent reconstitution into different formats such as hollow spheres, fibers, and films was achieved. Different proteins such as the green fluorescent protein or IgG binding domains GB1 or Z derived from protein G or protein A, respectively, were implemented to monitor the retention of protein function upon generation of reformatted materials. Materials were characterized and the retention of protein functionality was studied by assessing the fluorescence or IgG binding capacity. Since the Z domain protein functionality is retained, it suggests that protein refolding properties are critical parameters for restructuring these functional materials. This study shows that bioengineered biologically assembled protein-coated biopolymer particles can be completely dissolved and reformed into fibers, films, and hollow spheres retaining the original protein function.

Multiplexed Detection, Partitioning, and Persistence of Wild-Type and Vaccine Strains of Measles, Mumps, and Rubella Viruses in Wastewater.

Wastewater surveillance of vaccine-preventable diseases may provide early warning of outbreaks and identify areas to target for immunization. To advance wastewater monitoring of measles, mumps, and rubella viruses, we developed and validated a multiplexed RT-ddPCR assay for the detection of their RNA. Because the measles-mumps-rubella (MMR) vaccine is an attenuated live virus vaccine, we also developed an assay that distinguishes between wild-type and vaccine strains of measles in wastewater and validated it using a wastewater sample collected from a facility with an active measles outbreak. We also evaluated the partitioning behavior of the viruses between the liquid and solid fractions of influent wastewater. We found that assaying the liquid fraction of the wastewater resulted in more sensitive detection of the viruses despite the fact that the viral RNA was enriched in the solid fraction due to the low solids content of the influent wastewater. Finally, we investigated the stability of measles, mumps, and rubella RNA in wastewater samples spiked with viruses over 28 days at two different concentrations and two temperatures (4 °C and room temperature) and through freeze-thaw and observed limited viral decay. Our study supports the feasibility of wastewater monitoring for measles, mumps, and rubella viruses for population-level surveillance.
